Report on the Assessment of Contact Hypersensitivity of lsophorone Diisocyanate in Female Albino in Hartley Strain Guinea Pigs (CAS No. 4098-71-9)

Abstract

lsophorone diisocyanate was evaluated for. its potential as a sensitizing agent for contact hypersensitivity in guinea pigs. IPDI is used as an intermediate for polyurethanes with high stability, resistant to light discoloration and chemical attack. Other uses include as a raw material in paints, varnishes and elastomers.

The objective of this study was to determine the sensitizing potential of isophorone diisocyanate when applied to female albino (Hartley strain) guinea pigs.

Isophorone diisocyanate was tested on female albino (Hartley strain) guinea pigs. The doses of isophorone diisocyanate ranged from 0.1% to 1.0% in a solution of 4 parts acetone to one part olive oil (4+11) for sensitization and 3.0% for challenge. Guinea pigs received 100 µl by direct dermal application for 14 consecutive days to a site prepared by shaving and dermabrasion. Hexamethylene diisocyanate was used as a positive control at a concentration of 2.0% for sensitization and 0.1% for challenge. Measurement of the contact hypersensitivity response was done by visual evaluation (scoring).

lsophorone diisocyanate produced a dose-dependent irritation and a dose dependent contact hypersensitivity response in female albino (Hartley strain) guinea pigs.
